The land that would eventually become Sikeston was privately owned for many decades after the Louisiana Purchase until John Sikes purchased it and started planning the city.

This area was desirable because it sat at the intersection of a major road and a railroad, making it ideal for trade. During the Civil War, Sikeston was strategically very important because of its position on the highway.

Both Union and Confederate troops made their way through Sikeston at various points throughout the war, although neither of them ever stopped there. The city grew very quickly after the war because an early railroad terminated there.

In the 20th century, the town organized a local infantry company, which served in both World War I and World War II. The city also built a small airport in the 1930s.

The population continued to grow up until the 1990s, but has since slowly gotten smaller. While the city is connected to a large freight railroad network, there is no longer any passenger rail in the area.
